    CJ-023,423 (Grapiprant) a potential novel active compound with antihyperalgetic properties for veterinary patients

    Companion animals are now living longer and so are more commonly manifesting age- and pain-related disease. Nonsteroidal antiinflammatory drugs are the most used drug in osteoarthritis and inflammatory pain of various aetiologies. Despite their safety profiles have been amended from the COX-non selective to the COX-2 selective inhibitor class, some adverse effects are still of concern especially in long term treatments. One prostaglandin (PG) downstream from the cyclooxygenase enzyme, PGE2, has been recognized as a pivotal mediator of pain and inflammation. The actions of PGE2 are produced by its interaction with four G-protein coupled receptors (EP1, EP2, EP3 and EP4). The EP4 receptor mediates PGE2-elicited sensitization of sensory neurons and studies have demonstrated that EP4 is a major receptor in mediating pain associated with both rheumatoid and osteoarthritis and in inflammation. CJ-023,423 (grapiprant) is a competitive antagonist of human and rat prostanoid EP4 receptors, under development for the control of pain and inflammation associated with osteoarthritisfor use in humans and dogs. A recent study has shown the good safety profile of this active ingredient in dogs. Despite this molecule is still far to be marketed because it pharmacokinetic/pharmacodynamics profile is need to be fully elucidated yet, it might be an interesting active ingredient for the veterinary medicin

    Mechanobiological predictions of fetal joint morphogenesis

    This PhD thesis explores, through the use of a mechanobiological simulation of prenatal joint morphogenesis, the hypotheses on how fetal movements, shapes and position impact on the shape of the developing joint. A novel mechanoregulation algorithm specific for cartilage growth was developed and, for the first time, a 3D mechanobiological simulation of joint morphogenesis in which the effects of a range of movements and different initial joint shapes was proposed. Both pre- and post-cavitational phases of joint development were simulated and the effect of rigid paralysis on joint shape was also explored. This study concluded that the starting joint configuration and applied movement are fundamental for the development of specific and anatomically recognisable joint shapes. Moreover, for the first time, a mechanobiological simulation of prenatal hip joint morphogenesis was used to investigated the effects of reduced, or asymmetric, movement at various stages of fetal hip joint development. This study concluded that normal fetal movements are important for the emergence of a physiological hip joint shape and that movements during development tend to minimise the natural trend of decreasing stability. Results showed that reduced movements at an early stage of development lead to decreased sphericity and acetabular coverage of the femoral head, increasing the risk of subluxation or dislocation of the hip. It also shows that, in the case of mal-positioning or joint laxity in utero, movements may actually lead to an abnormal hip joint shape with characteristics of developmental dysplasia of the hip (DDH). This PhD thesis has advanced the basic understanding of prenatal joint shape development and the implication that different mechanical environments within the joint region, might have on developmental skeletal diseases such as DDH.Open Acces

    The university radio in Argentina. The case of the station of the National University of Avellaneda

    Este artículo aborda la historia y la situación de la radio universitaria argentina en la actualidad, los aspectos de la programación de estas emisoras, su vínculo con Internet y las nuevas tecnologías, y su desarrollo en Argentina a partir del trabajo en una red nacional de radiodifusoras, recuperando las principales aportaciones teóricas en este sentido. Posteriormente, el trabajo se enfoca en el estudio de caso de Radio UNDAV, la emisora de la Universidad Nacional de Avellaneda, Argentina, puesta en marcha en el año 2011. En este sentido, se da cuenta de la experiencia de esta emisora en torno a los aspectos teóricos abordados en la primera parte del artículo y a partir de un estudio documental, un análisis de la programación, encuestas a integrantes de los programas y una recopilación de los mecanismos de articulación académica implementados. Finalmente, se exponen a modo de conclusión, una serie de premisas en torno a los resultados obtenidos.This article addresses the history and situation of Argentine university radio today, the aspects of the programming of these stations, their link to the Internet and new technologies, and their development in Argentina through work on a national network of radio broadcasters , recovering the main theoretical contributions in this sense. Subsequently, the work focuses on the case study of Radio UNDAV, the station of the National University of Avellaneda, Argentina, launched in 2011. In this sense, it is reported the experience of this station around the theoretical aspects addressed in the first part of the article and from a documentary study, an analysis of programming, surveys of program members and a compilation of the mechanisms of academic articulation implemented. Finally, a series of premises based on the results obtained are presented as a conclusion

    Trazodone: A review of its pharmacological properties and its off-label use in dogs and cats

    Trazodone is approved by the Food and Drug Administration (FDA) for the treatment of depression and anxiety in humans, it can perform these functions due to its selective serotonin receptor antagonism and reuptake inhibition properties. Trazodone has not been approved for animals by the FDA but it is legally prescribed by veterinarians as an off-label drug for dogs and cats. This review describes trazodoneâ\u80\u99s mechanism of action, pharmacokinetic properties and adverse effects and summarizes the benefits of trazodone for non-approved FDA indications to reduce anxiety, behavioral stress, sedation, pre-and post-operation stress. It also discusses its cardiac safety profile for dogs and cats. As few clinical trials have been conducted to evaluate trazodoneâ\u80\u99s efficacy for treatment of the many behavioral and medical conditions in animals, large and randomized controlled clinical trials are still needed to confirm its efficacy in clinical practice
